Egyptian hieroglyphic inscriptions dating to approximately 2641 BCE described the sudden death of Pharaoh Menes, believed to have been caused by a wasp or hornet sting, representing one of the earliest possible recorded cases of fatal anaphylactic reaction. Ancient physicians across Greek, Roman, and Chinese traditions noted that certain substances caused swift and dramatic systemic responses in some individuals, though no conceptual framework existed to explain why similar exposures proved harmless to most and catastrophic to a few.

Hippocratic and Galenic medical traditions attributed sudden systemic collapses to the rapid corruption of humors or the overwhelming of vital spirits by toxic substances. The concept that the body's own defensive mechanisms could turn destructive was entirely absent from ancient and medieval thinking, which generally understood physiological responses as either protective or failing rather than as paradoxically harmful.

For much of the medieval and early modern period, cases resembling anaphylaxis were interpreted through the lens of poison, demonic influence, or constitutional weakness. Physicians documented sudden deaths following bee stings, certain foods, and medicinal preparations, recording these events as medical curiosities without theoretical frameworks to connect them.

The formal scientific discovery of anaphylaxis came in 1902 through the collaborative work of Charles Richet and Paul Portier aboard the yacht of Prince Albert I of Monaco. The researchers were attempting to understand why certain marine toxins caused such violent reactions in humans, hoping to develop a protective immunization against jellyfish and sea anemone venoms. When they injected dogs with small, non-lethal doses of these toxins and returned days later to administer a second dose, they expected to find the animals had developed protection. Instead, many dogs died within minutes of the second injection, demonstrating that prior exposure had somehow increased rather than decreased sensitivity. Richet coined the term anaphylaxis in 1902, combining the Greek prefix meaning against with the word for protection, to describe this paradox of immunity working in reverse.

Richet was awarded the Nobel Prize in Physiology or Medicine in 1913 in recognition of this discovery, which opened entirely new lines of inquiry into immunological hypersensitivity. His work prompted other researchers to investigate whether similar mechanisms might explain hay fever, asthma, and other conditions that had long puzzled physicians.

Nicholas Maurice Arthus independently contributed to understanding hypersensitivity reactions in 1903 when he described a localized form of intense inflammatory reaction following repeated subcutaneous injections in rabbits, a phenomenon that became known as the Arthus reaction and provided a parallel model for understanding immune hypersensitivity.

In the decades following Richet's discovery, researchers including Clemens von Pirquet and Béla Schick refined the concept of altered immunological reactivity, introducing the term allergy in 1906 to describe this broader category of aberrant immune responses. These contributions gradually built the theoretical foundation that later investigators would use to identify specific mediators responsible for the physiological cascade observed during severe hypersensitivity reactions.
