Scholars have argued that certain passages in the Hebrew Bible, particularly those describing one of the plagues of Egypt involving festering boils and the sudden death of livestock, may represent early accounts of anthrax outbreaks. Ancient Hindu texts in the Atharvaveda described a fatal disease of cattle exhibiting features consistent with anthrax, and Roman agricultural writers including Virgil and Varro wrote about a pestilence that periodically swept through flocks and herds, leaving animals dead with darkened blood, a characteristic the disease's Greek-derived name would eventually commemorate.

The word anthrax itself derived from the Greek word for coal, a reference to the dark eschar that formed on the skin of affected individuals. Ancient Greek physicians including Hippocrates and later Galen recognized a disease of this name, and the Hippocratic writings described a malignant pustule producing blackened tissue. Medieval physicians working within the Galenic tradition attributed such lesions to an excess of black bile and prescribed cauterization, bloodletting, and herbal poultices to draw out the corrupting humors. The German physician Johann Georg Adam Forster contributed early descriptive accounts, and by the early nineteenth century European veterinary schools had begun systematic study of the disease in animals. In the 1840s and 1850s, German physician Aloys Pollender and French physician Casimir Davaine independently observed rod-shaped bodies in the blood of anthrax-killed animals under the microscope, though their findings awaited proper interpretation.

The decisive breakthrough came through the work of Robert Koch, who in 1876 published his rigorous experimental demonstration that Bacillus anthracis caused anthrax, tracing the entire life cycle of the bacterium and identifying its spore-forming capacity. Koch's anthrax work established methodological standards for proving microbial causation that became foundational to all subsequent bacteriology. Louis Pasteur, Koch's great rival, contributed the next landmark achievement when in 1881 he staged his famous public demonstration at Pouilly-le-Fort, vaccinating sheep, goats, and cattle against anthrax with an attenuated bacterial preparation and demonstrating dramatic protection against a subsequent challenge dose before an audience of scientists, journalists, and government officials. Pasteur's triumph was a watershed moment in the history of vaccination and transformed public confidence in the nascent science of immunology.

Throughout the late nineteenth and early twentieth centuries, anthrax remained an occupational hazard among wool sorters, hide handlers, and tannery workers, and industrial epidemics in England's wool districts gave rise to the colloquial name woolsorter's disease for pulmonary cases. Researchers in this period worked to understand how different routes of exposure produced different clinical patterns, advancing knowledge of infectious disease pathophysiology.
