Before the bacteriological era, what would later be identified as melioidosis almost certainly afflicted people in tropical regions of Asia and beyond without any distinct recognition. Conditions involving severe fever, abscesses, and systemic illness in tropical climates were grouped under vague categories in ancient and medieval medicine, attributed variously to miasmatic air, excess heat and moisture acting on the humors, or divine disfavor. Classical Indian texts described febrile illnesses with suppurative features in tropical settings, but no specific identification of melioidosis as an entity can be drawn from these sources with confidence.

The formal discovery of the causative organism is attributed to Alfred Whitmore, a British army pathologist stationed in Rangoon, Burma, who in 1911 and 1912 investigated a cluster of deaths among morphine addicts and other patients at the Rangoon General Hospital. Whitmore and his colleague C.S. Krishnaswami isolated an unusual bacterium from postmortem tissues and published their findings in 1912, describing a new disease characterized by glanders-like lesions and caused by an organism they called Bacillus pseudomallei, with the prefix pseudo reflecting their observation of its resemblance to the glanders bacillus. The term melioidosis itself, derived from the Greek for a distemper of asses combined with the suffix for resemblance, was proposed to capture this relationship to the well-known equine and human disease glanders.

In the decades following Whitmore's discovery, scattered case reports emerged from Southeast Asia, Australia, and other tropical regions, though the condition remained poorly known outside specialist circles of tropical medicine. Arthur Stanton and William Fletcher conducted further investigations in Malaya in the 1920s, contributing to the early microbiological characterization of the organism and documenting its presence in soil and water, which hinted at an environmental reservoir even before its ecology was fully understood.

World War II brought melioidosis to greater medical attention as Allied forces campaigned extensively in Southeast Asia and encountered the disease among troops. Military medical officers documented cases and struggled to identify the condition amid the many febrile illnesses afflicting soldiers in tropical environments. The Vietnam War era in the 1960s and early 1970s further elevated the condition's profile in Western medical awareness, as American military physicians encountered cases among troops deployed to Southeast Asia and the condition acquired the informal wartime designation of the 'Vietnamese time bomb' due to its propensity to reactivate after long dormant periods. This military medical experience stimulated research and led to greater engagement with the organism by microbiologists and infectious disease specialists.

The reclassification of the causative organism from its original Bacillus and later Pseudomonas genus designations to the newly established genus Burkholderia in 1992 reflected decades of evolving bacteriological understanding and represented the culmination of extensive taxonomic and genetic research by scientists including Yabuuchi and colleagues. This reclassification situated Burkholderia pseudomallei within a broader group of environmentally important bacteria and opened new avenues of scientific investigation into the organism's biology and the history of the disease it caused.
